Penicillin fermentation and role of strain improvement on efficiency Introduction Penicillin is one of the most vital drugs known to the medical world. Its discovery by Alexander Fleming in late 1920s changed the face of the world by bringing hope in peoples lives as penicillin was effectively used against diseases such as syphilis and staphylococcus infections. Penicillins discovery was an unexpected accident carried out while isolating staphylococcus aureus by growing bacteria on petri dishes. It was discovered that on one of the dishes a contaminating mould named penicillium notatum had no bacteria around it. Fleming noticed this behavior and obtained a small amount of the secreted antimicrobial product and named it penicillin. However Fleming was not able to commercialize the product into starting a large scale production process and preserved the cultured organism. As penicillin became more popular among other researchers, fermentation processes were being developed to improve its yield. [3]. Using these fermentation techniques, researchers started to carry o ut the production of penicillin in labs at a smaller scale. This process required a considerable amount of time and effort. One of the first methods used to produce penicillin came not too soon after its discovery. The first production procedure resulted in penicillin being produced by fermentation using surface method. In this method, penicillium mould was grown on top of a quiescent medium and bottle plant techniques were used to yield penicillin. This was a descent breakthrough in producing the antibiotic drug but it took very long growing cycles of penicillium mould. [3]. The demand for antibiotics really went up during and after the Second World War. At this time penicillin was well known around the world, however its production at larger scale with better efficiency was needed to make it a household name. The challenge of producing penicillin at larger scale was very daunting for researchers and engineers. It was at that time when fermentation research on corn steep liquor at a laboratory in Illinois, USA allowed in producing 2.3 million doses through development of deep tank fermentation [5]. As the demand for penicillin was rising, different ways of obtaining the antibiotic drug increased. Such production methods involved the use of fermenters with buffers and separation funnels, the property of liquid-liquid extraction, distribution ratios and finally the use of a membrane in a countercurrent extraction column. Penicillin Production Procedures Since the discovery of penicillin as an antibiotic drug, there have been many different methods developed to produce penicillin efficiently in order to achieve maximum yield. For instance, the recovery of penicillin using an emulsion liquid membrane in countercurrent extraction column is very successful method yield a highly qualitative product. The use of a countercurrent extraction column requires both a dispersed phase and an aqueous phase. The aqueous phase is made of penicillin potassium salt which is dissolved in a citrate buffer solution while the dispersed phase is made of a mixture of internal aqueous solution; sodium carbonate present in de-ionized water and organic solution, a mixture of a secondary amine, and nonionic polyamine in kerosene [1]. In order to process this method, the continuous phase is first fed through the top of the column at unsteady state, and when the flow of this phase reaches steady state, the dispersed phase is injected through the bottom of the col umn, producing a countercurrent extraction with the help of a nozzle. While the process is extracting, time samples are taken from the top and the bottom to monitor the concentration of penicillin. Once the separation of the continuous phase and dispersed phase occurs with the help of a filtration process, the penicillin that was present in the continuous phase is recovered. The effectiveness of the removal of different types of penicillin can be removed at a greater value at lower pH. This is because at a lower pH the impurities are left behind and can be removed, but at a higher pH, the impurities will remain with the organic phase, and thus will be hard to separate. This is the major reason why the aqueous extract is acidified again, so that we are able to lower the pH and obtain a better amount of penicillin and remove most of the impurities present [4]. Although methods like Emulsion liquid membrane in countercurrent extraction column can be very productive and economically feasible, some other measures are still required to produce penicillin in industry at a larger scale. Industrial Procedure The industrial process of the recovery of penicillin is in fact more complex. It requires the use of a fermenter, a filtration system and 3 buffer units, as shown in figure below. The industrial process uses the same principle of distribution ratio, which determines the measure of purity in the extracted product. The distribution ration is equal to the concentration of a solute phase in an organic phase divided by its concentration in aqueous phase. This principle is effectively used in removing the level of impurities from the final product. In this process, corn-steep liquor, which is a waste product in the wet milling of corn, is entered as the feed. When it enters the fermenter, additional nutrients such as metabolism modifiers and antifoaming agents can also be added. The pH is then adjusted and the contents in the fermenter are seam sterilized. As the temperature reaches a value of about 25oC, the fermenter is inoculated with a pure and high yielding mutant strain of Penicillium [2]. At this point, it comes in contact with sterile air with the help of turbine agitation to provide proper air medium interface and the constant addition of extra nutrients an d antifoaming agents [2]. This whole process is carried out for approximately one week. After the fermentation period, the solution is then sent to a filtration unit where impurities such as mycelium are separated from the aqueous solution that contains penicillin. As the separation takes place, the aqueous solution goes through three extraction units to obtain the final product in solid form. First the solution enters the primary extraction unit where its pH is reduced to a value of two by using sulfuric acid and amyl acetate, these results in penicillin to be extracted. This organic solution is then sent into the secondary extraction unit where it comes into contact with an aqueous buffer solution with a pH of six. This results in the formation of rich penicillin aqueous solution. Finally this solution is sent to the third and final extraction unit. Here the solution is re-acidified and either contacted with the same organic solvent used in primary extraction or with another organ ic solvent such as methyl isobutyl ketone. This second contact with organic solvent is used to prevent the transfer of any soluble impurities [2]. After continuous treatment, the final product of penicillin is recovered in solid form. Conclusion Penicillin is world renown antibiotic, its importance came in the early 1900s were it was used as a healing agent for bacterial infections. Till today its importance plays a vital role in society. Penecillin process is carried out with the help of new ways being developed in the recovery of penicillin. To conclude, it was noted that there are many different ways to recover penicillin using different agents. However, the quality and quantity of the product plays an important role in determining the best fermenting process. Using the principles of distribution ratios recovered penicillin in an almost pure form. Lesson Learned There are so many events that change one’s life that it is rather difficult to try and decipher which of those events are most important. Each event changes a different aspect of your life, molding how one’s personality turns out. One of these events occurred when I was about twelve years old and I attempted to steal from a Six Flags amusement park. My reasoning for stealing wasn’t that I didn’t have the money, or even that I wanted what I stole all that badly, it was that all of my friends had stolen something earlier that day and didn’t get caught. After getting caught I resolved, because the consequences are just not worth it, never to steal or give into peer pressure again. It all began when a couple of friends and myself went to Six Flags for some fun right at the beginning of summer. We arrived there early in morning, which was about nine o’clock for us. The birds were chirping and the sun was bright. There was also a hint of dew, making the grass sparkle in the sunlight. The place was packed, people were standing in huge lines waiting to buy their tickets. My friends and I were lucky because we had season passes and just walked right to the front gate and walked in. Naturally there was a security check at the front gate, little did I know that I was going to run into them again in the near future. From there we went straight to the ride â€Å"The Mind Eraser.† It was always wise to go to that ride first, because as the day progressed, the line grew longer. Since the ride is only about a minute long, the wait then becomes to long to make the ride worth the wait. As we got on, all of us were a little jumpy. It was the first time being to the amusement park that year, and our stomachs were not immune to the s... ...o our red van not saying a word to me. On the drive home she gave me my punishment. The punishment was that I was to be grounded for an entire month, and reap the consequences with the law. I ended up sitting in my house for the first month of summer and losing two-hundred and eighty dollars, which was my fine for stealing. After that event, I made things very clear what I would never do again. From then on I never did something just because everyone else did. I would assess the situation and decide whether the action was worth it’s consequences. This event prevented me from becoming one of those people who break the law just because they can, or because it gives them an adrenalin rush. My actions on that fateful day were definitely not worth the consequences that I suffered, and because of that, I have been very obedient of the laws and rules that are around me.

They offer many developmental classes, schools and academies that give you the tools to become a successful leader. Like most jobs there is a pyramid effect of leadership roles. This pyramid role is called a chain of command in the military. Each leadership role is just as important as the one appointed above them and is only as strong as its weakest link.There are several types of leaders that dictate our military; authoritative, hands-on and hands-off. Each type of leader possesses different qualities which make the outcome of their work diverse. Authoritative leaders are born to be a leader. These are leaders who have a natural instinct on how to take control of a group and get the job done. They are the type of person you either love or hate. Authoritative leaders are very meticulous and strict on how they want to accomplish a task. It’s either their way or the highway. This can be very draining on the soldiers that work for them.Authoritative leaders can take two separate approaches in leading their soldiers. They either let the power get to their heads and bark orders killing the moral of the soldiers they work with. The other approach of authoritative leadership is someone who delegates work by asking for help on what needs to be accomplished, making it a much more comfortable environment to work in. These separate approaches can dictate the outcome of the work being performed by their soldiers which can either make or break their leadership role.Hands-on leaders are one of the most effective leaders in the military. These are leaders who don’t mind getting their hands dirty and take initiative in getting the job done. They are leaders who want to lead by example and are not afraid to take the blame if something goes wrong. Hands-on leaders are very well communicator’s, open to su ggestions and look for soldiers input. This allows them to expand not only their own knowledge but also the soldiers around them. They are willing to work with their soldiers on the small things in order to accomplish something larger.This gives them a better understanding of their job and the soldiers they work with. Once a hands-on leader sets the goals and shows the soldiers what they expect, it allows them to take a step back and let their soldiers execute the tasks at hand. Hands-off or laissez-faire leaders are considered the lazy one’s of the group. They typically are the fun leaders to hang out with as a friend but lack leadership as a boss. Hands-off leaders take no guidance of the group and are very passive on work. They tend to pass off jobs and allow the soldiers to dictate how the work is going to be done.This can either set the soldiers up for success or can completely kill the mission. Hand-off leaders are considered the chameleon of the group. They take some o f the credit on a successful task or hide and push the blame onto someone else when the task fails. Hands-off leaders typically try to please everyone to avoid any conflict. This can ultimately hurt a unit by not being able to trust or rely on their leaders. There are many routes a leader can take in leading their soldiers. It is up to the soldier to decide what leader he or she wants to become.Every day the military is looking for someone to step up and take on one of the roles of becoming a leader. The best way to decide on what leader you want to become is to look around and take notes on the qualities your leader’s posses and make a decision on how you want your peers to perceive you. Each leadership role can create a different outcome that can either take you straight to the top for success or damage your chances of becoming a great leader. It is up to you to make the decision on what path you want to take and the leader you would like to become.

Over the past 25 years, no place has been more perilous for companies than Colombia, a country that is finally beginning to emerge from the effects its Colombian banana subsidiaries had made protection payments to terrorist groups from 1997 through 2004. The Justice Department began an investigation, focusing on the role and conduct of Chiquita and some of its officers in this criminal activity. Subsequently, Chiquita entered into a plea agreement that gave them the dubious distinction of being the first major U. S. company ever convicted of dealing with terrorists, and resulted in a fine of US$25 million and other penalties. To make matters worse, the industry was facing pressure from increasing retailer purchasing power, major changes in consumer tastes and preferences, and Europe’s imposition of an â€Å"onerous tariff† on companies that sourced bananas from Latin America. Chiquita’s top m anagement team wasn’t making decisions in unison and you could tell this by the various executives either quitting or retiring. The guilt from the different stances each executive took made them eventually leave the company because they had dug themselves as well as the company into the ground.I also believe the cause for Chiquita to get convicted was their values and ethics. I believe this because in the case, Chiquita tried to change their core values in 1999 to better the company. If they believed in strong values and ethics from the start, they would have never had to revamp their core values. Chiquita should’ve taken charge from the start with their decision making styles and their values and ethics so that they would have never placed themselves in the position to pay the AUC which in turn led to their conviction.With a sound team with sound goals, Chiquita will always prosper internally. I believe Chiquita and its managers did have a choice in the AUC payments. Yes Chiquita was doing business in an unstable political environment but they should have taken a stance and fought for what they thought was right. The actions the manager partook in were not ethical principles and I believe they were looking out more so for themselves than the company as a whole. I believe Chiquita could have came up with a strategy that would have made both the company profitable and satisfy its stakeholders without paying the AUC.If Chiquita felt like it didn’t have a choice, they should have involved the law enforcements well before they started the payments. If they would have done this, they would have never had to worry about the safety of its company or the terrorist groups in Colombia. Other companies that should be worried about Chiquita’s experience are companies that were involved in similar circumstances as Chiquita. In the case, it stated that Chiquita was the first company to get convicted for paying terrorist groups.If similar companie s were doing the same, they should cease the payments and learn from Chiquita’s mistakes because it stated that the law was cracking down and getting harder on this crime. Companies that are doing business in lesser developed economies should keep the employees’ safety at heart but at the same time do business in ethical ways. The factors of the political and economic environment should also be in concern when doing business in these lesser developed economies so that they will make sound business decisions.This story changes my perspective about doing business abroad because it shows me that just because you have a competitive advantage and resources to make millions doesn’t mean you have a safe environment to run your business in. It helps me understand that there is more to business than making money and that you have to have all of your business standards and strategies in place to successfully operate a company abroad. The current CEO can implement the stra tegies I listed above in my strategic analysis to restore Chiquita’s reputation and ensure future competitiveness.With the implementation of these strategies, Fernando can lead the company in the right direction and provide a stronger stance for the company. The main thing for the company to do is to revamp its mission and vision statement because this is the most important start for the company. The admission ticket serves as proof that you are registered to take the ACT on a given day. Food is flying everywhere, lettuce is being launched through the air, and there are pieces of meat all over the table and floor. You are astounded to see that he is trying to snag his third burger. You then walk away utterly shocked that someone can let their dog get that fat. That is right what I was describing was about a canine, not a human being, which is what I bet you thought while reading the description. Society and the media have put this picture into our heads that obese people sit at restaurants all day stuffing their faces with food, and are extremely lazy. As it turns out, this depiction of obese people is wrong, and there is more involved in obesity than you think. There are a hundred causes as to why a person is obese, but thanks to society and the media we only think of the one reason. In fact, some of the common causes of obesity are diseases. There are many syndromes that could cause obesity but the most common ones are hypothyroidism, Cushing’s disease, Prader-Willi syndrome, and polycystic ovarian syndrome. As society frequently passes judgment on obese people they do not stop to think that a disease could be the cause for their obesity. The first genetic disorder that is involved in causing obesity is called hypothyroidism. This disease is fairly common as it affects more than forty percent of Americans (Lowrance, 2009). In order to explain how this disease causes obesity, I will first explain what t... ...ypothyroidism, which is caused by an underactive thyroid gland. Fifteen million have a disease called Cushing’s disease, which happens when the body is overexposed to cortisol. Prader-Willi affects thirty thousand, and it caused by a bad fifteenth chromosome. Fifty percent of women who have polycystic ovarian syndrome are obese. This condition is caused by â€Å"insulin resistance and biochemical signaling.† All of these people affected by these disorders are obese because of something that they could not control, genetics, not overeating and laziness. So please, next time you see an overly obese person think first before you judge them. The quote said by Adelle Davis perfectly sums up my argument, â€Å" To say that obesity is caused by merely consuming too many calories is like saying that the only cause of the American Revolution was the Boston Tea Party,† (Davis, n.d).
